The precise mechanism by which Lacosamide exerts its antiepileptic effects in humans remains to be fully elucidated
The precise mechanism by which Lacosamide exerts its antiepileptic effects in humans remains to be fully elucidated. In vitro electrophysiological studies have shown that lacosamide selectively enhances slow inactivation of voltage-gated sodium channels, resulting in stabilization of hyperexcitable neuronal membranes and inhibition of repetitive neuronal firing. Lacosamide binds to collapsin response mediator protein-2 (CRMP-2), a phosphoprotein which is mainly expressed in the nervous system and is involved in neuronal differentiation and control of axonal doutgrowth. The role of CRMP-2 binding in seizure control is unknown.
Purpose
Epilepsy
Ingredients
Lacosamide
Warnings
Treatment with lacosamide has been associated with dizziness which could increase the occurrence of accidental injury or falls.
Prolongations in PR interval with lacosamide have been observed in clinical studies. Lacosamide is advised to be used with caution in patients with known conduction problems or severe cardiac disease such as a history of myocardial infarction or heart failure.
Lacosamide is not recommended for use in children & adolescents below the age of 16. The safety and effectiveness of Lacosamide in pediatric patients <17 years have not been established.
Side Effects
The adverse events most commonly (>1% in the Lacosamide total group and greater than placebo) leading to discontinuation were dizziness, ataxia, vomiting, diplopia, nausea, vertigo, and vision blurred.
