The primary site of action of rosuvastatin is the liver, the target organ for cholesterol lowering. Rosuvastatin increases the number of hepatic LDL receptors on the cell-surface, enhancing uptake and catabolism of LDL and it inhibits the hepatic synthesis of VLDL, thereby reducing the total number of VLDL and LDL particles.

Warnings
Rosuvastatin is advised to be used with caution in patients who consume substantial quantities of alcohol and/or have a history of chronic liver disease.
A dose reduction is advised to be considered for patients on rosuvastatin therapy with unexplained persistent proteinuria and/or hematuria during routine urinalysis testing.
Rosuvastatin is advised to be prescribed with caution in patients with predisposing factors for myopathy/rhabdomyolysis. . Such factors include: renal insufficiency, hypothyroidism, personal or family history ofhereditary muscular disorders, previous history of muscular toxicity with another HMG-CoA reductase inhibitor or fibrate, alcohol abuse.
Side Effects
Headache, dizziness, constipation, nausea, abdominal pain, myalgia, asthenia, diabetic mellitus Pruritus, rash and urticaria , Hypersensitivity reactions including angioedema, myopathy, rhabdomyolysis, pancreatitis.
